Fed-batch culture of hybridoma cells in serum-free medium using an optimized feeding strategy

چکیده
The achievement of high cell density and monoclonal antibody concentration depends on understanding cell metabolism and the control of cell culture. A model of cell metabolism and monoclonal antibody production in hybridoma cell culture is presented. Fed-batch culture in serum-free medium was conducted in a bioreactor with an optimized feeding strategy according to the model. In the process, nutrient concentrations, especially glucose and glutamine, were maintained at relatively constant and low levels. The formation of toxic byproducts, such as ammonia and lactate, was greatly reduced in comparison with that in non-optimized batch culture. The concentration of monoclonal antibody reached 350 mg dm−3, which was seven times greater than in non-optimized batch culture. 2004 Society of Chemical Industry

High cell density and high monoclonal antibody production through medium design and rational control in a bioreactor.
A simple feeding strategy was developed and successfully employed for nutritional control in a 2-L fed-batch culture of hybridoma cells. A previously developed stoichiometric model for animal cell growth was used to design a supplemental medium for feeding. Undialyzed fetal bovine serum and trace metals (Fe(2+), SeO(3) (2-), Li(+), Zn(2+), and Cu(2+)) were fed to the cells periodically in addit...
متن کاملOptimized fermentation conditions for improved antibody yield in hybridoma cells
Background Traditionally antibody producing cells like hybridoma cells sank into oblivion since other suspension cell lines have captured the biopharmaceutical production market. However, they are still of particular interest in academic and industrial diagnostic research. Hence, fast and sufficient antibody production is needed as proof of concept, for toxicology and in vivo studies. Although,...
متن کامل-Amylase Production in Fed Batch Cultivation of Bacillus caldolyticus: An Interpretation of Fermentation Course Using 2-D Gel Electrophoresis
The conditions for increased production of thermostable -amylase from Bacillus caldolyticus DSM 405 were investigated. Preliminary experiments in batch shake flasks led to an optimized initial cultivation medium. Shake flask experiments in extended-batch and in fed-batch mode of operation indicated that the -amylase production was enhanced by continuous feeding of starch. The activity of the -a...
متن کاملPopulation balance between producing and nonproducing hybridoma clones is very sensitive to serum level, state of inoculum, and medium composition.
Secreting and nonsecreting hybridoma populations derived from the murine hybridoma cell line 167.4G5.3 were each grown in batch culture in low serum and serum-free media. Under serum-free conditions, a secreting population gained on a predominantly nonsecreting population and competed with the existing antibody-deficient cells effectively. It was found that this competition was sensitive to sta...

Researchers add serum to a classical medium at concentrations of 5 to 10% (v/v) to grow cellsin-vitro culture media. Unfortunately, serum is a poorly defined culture medium componentas its composition can vary considerably while serum-free cell culture media are an excellentalternative to standard serum-containing media and offer several major advantages.
